{"id":15053,"date":"2018-03-08T08:21:59","date_gmt":"2018-03-08T08:21:59","guid":{"rendered":"http:\/\/wames.org.uk\/cms-english\/?p=15053"},"modified":"2018-03-08T08:25:49","modified_gmt":"2018-03-08T08:25:49","slug":"investigation-of-mast-cell-toll-like-receptor-3-in-cfs-me","status":"publish","type":"post","link":"https:\/\/wames.org.uk\/cms-english\/investigation-of-mast-cell-toll-like-receptor-3-in-cfs-me\/","title":{"rendered":"Investigation of mast cell toll-like receptor 3 in CFS\/ME"},"content":{"rendered":"<h3><strong>Research abstract:<\/strong><\/h3>\n<p><a href=\"http:\/\/apjai-journal.org\/wp-content\/uploads\/2017\/12\/AP-200517-0086.pdf\" target=\"_blank\" rel=\"noopener\">Investigation of mast cell toll-like receptor 3 in Chronic Fatigue Syndrome\/Myalgic Encephalomyelitis and Systemic Mastocytosis participants using the novel application of autoMACS magnetic separation and flow cytometry<\/a>, by C Balinas, T Nguyen, S\u00a0Johnston, P\u00a0Smith, D\u00a0Staines, S\u00a0Marshall-Gradisnik <span style=\"text-decoration: underline;\">in<\/span> <em>Asian Pac J Allergy Immunol<\/em>. 2017 Dec 10 [Epub ahead of print]<\/p>\n<p>BACKGROUND:<\/p>\n<p><img data-recalc-dims=\"1\" decoding=\"async\" class=\"alignright size-medium wp-image-15793 lazyload\" data-src=\"https:\/\/i0.wp.com\/wames.org.uk\/cms-english\/wp-content\/uploads\/2018\/03\/Mast-Cell-300x221.jpg?resize=300%2C221\" alt=\"\" width=\"300\" height=\"221\" data-srcset=\"https:\/\/i0.wp.com\/wames.org.uk\/cms-english\/wp-content\/uploads\/2018\/03\/Mast-Cell.jpg?resize=300%2C221&amp;ssl=1 300w, https:\/\/i0.wp.com\/wames.org.uk\/cms-english\/wp-content\/uploads\/2018\/03\/Mast-Cell.jpg?resize=150%2C111&amp;ssl=1 150w, https:\/\/i0.wp.com\/wames.org.uk\/cms-english\/wp-content\/uploads\/2018\/03\/Mast-Cell.jpg?w=457&amp;ssl=1 457w\" data-sizes=\"(max-width: 300px) 100vw, 300px\" src=\"data:image\/svg+xml;base64,PHN2ZyB3aWR0aD0iMSIgaGVpZ2h0PSIxIiB4bWxucz0iaHR0cDovL3d3dy53My5vcmcvMjAwMC9zdmciPjwvc3ZnPg==\" style=\"--smush-placeholder-width: 300px; --smush-placeholder-aspect-ratio: 300\/221;\" \/>Viral infections and hypersensitivities are commonly reported by Chronic Fatigue Syndrome\/Myalgic Encephalomyelitis (CFS\/ME) patients. <a href=\"https:\/\/en.wikipedia.org\/wiki\/Mast_cell\" target=\"_blank\" rel=\"noopener\">Mast Cells<\/a> (MC) uniquely mediate type 1 hypersensitivities and resolve viral infections via <a href=\"https:\/\/en.wikipedia.org\/wiki\/Toll-like_receptor\" target=\"_blank\" rel=\"noopener\">toll-like receptor<\/a> 3 (TLR3).<\/p>\n<p>OBJECTIVE:<\/p>\n<p>To characterise and compare mast cell progenitors (MCPs) in CFS\/ME participants with a known MC disorder, Systemic mastocytosis (SM), and secondly, to investigate the role of MC TLR3 in CFS\/ME participants following Polyinosinic:polycytidylic acid (Poly I:C) stimulation.<\/p>\n<p>METHODS:<\/p>\n<p>A total of 11 International Consensus Criteria defined CFS\/ME participants (40.42 \u00b1 10.31), 9 World Health Organisation defined systemic mastocytosis (SM) participants (47.00 \u00b1 10.37) and 12 healthy controls (HC) (36.36 \u00b1 9.88) were included. Following autoMACS magnetic separation, CD117+\/Lin-MCPs were stimulated with Poly I:C for 24hr. MCP purity (CD117 and Lin2), maturity (CD34 and Fc\u03b5RI), interaction receptors and ligands (CD154 and HLA-DR), and SM-specific (CD2 and CD25) markers were measured using flow cytometry.<\/p>\n<p>RESULTS:<\/p>\n<p>There was a significant decrease in HLA-DR+\/CD154- expression between CFS\/ME and SM groups pre and post Poly I:C stimulation.
