Pharmacological activation of AMPK and glucose uptake in cultured\u00a0human skeletal muscle cells from patients with ME\/CFS<\/a>, by\u00a0Audrey E Brown, Beth Dibnah, Emily Fisher, Julia L Newton, Mark Walker in<\/span>\u00a0Bioscience Reports<\/em> [Preprint\u00a0April 13, 2018]<\/p>\n

Background:<\/strong>
\nSkeletal muscle fatigue and post-exertional malaise are key symptoms of\u00a0Myalgic Encephalomyelitis ( ME\/CFS). We have previously shown that AMPK<\/a>\u00a0activation and glucose uptake<\/a> are impaired in primary human skeletal\u00a0muscle cell cultures derived from patients with ME\/CFS in response to\u00a0electrical pulse stimulation, a method which induces contraction of\u00a0muscle cells in vitro. The aim of this study was to assess if AMPK could
\nbe activated pharmacologically in ME\/CFS.<\/p>\n

Methods<\/strong>
\nPrimary skeletal muscle cell cultures<\/a> from patients with ME\/CFS and\u00a0healthy controls were treated with either metformin or 991. AMPK\u00a0activation was assessed by Western blot and glucose uptake measured.<\/p>\n

Results<\/strong>
\nBoth metformin<\/a> and 991 treatment [antifungal] significantly increased AMPK activation\u00a0and glucose uptake in muscle cell cultures from both controls and\u00a0ME\/CFS. Cellular ATP content was unaffected by treatment although ATP\u00a0content was significantly decreased in ME\/CFS compared to controls.<\/p>\n

Conclusions<\/strong>
\nPharmacological activation of AMPK can improve glucose uptake in muscle\u00a0cell cultures from patients with ME\/CFS. This suggests that the failure\u00a0of electrical pulse stimulation to activate AMPK in these muscle\u00a0cultures is due to a defect proximal to AMPK. Further work is required\u00a0to delineate the defect and determine whether pharmacological activation
\nof AMPK improves muscle function in patients with ME\/CFS.<\/p>\n
